چکیده

Vessels within the lymphocytic infiltrate in 161 examples of 24 different diseases have been studied using light and electron microscopy and histochemistry. Hitherto undescribed venules have been identified within lymphocyte aggregates which exhibit a structure, ultrastructure and cytochemistry previously believed to be restricted to the high endothelial venules of lymphoid tissues. The significance of these findings is discussed within the context of lymphocyte migration into abnormal tissue.

Stabilizing the VE-cadherin-catenin complex blocks leukocyte extravasation and vascular permeability.

To determine whether leukocytes need to open endothelial cell contacts during extravasation, we decided to generate mice with strongly stabilized endothelial junctions. To this end, we replaced VE-cadherin genetically by a VE-cadherin-α-catenin fusion construct. Such mice were completely resistant to the induction of vascular leaks by VEGF or histamine. Neutrophil or lymphocyte recruitment into...

Pertussis toxin inhibits migration of B and T lymphocytes into splenic white pulp cords

The normal migration route of B cells into follicular areas of spleen and lymph nodes is altered in the case of autoreactive cells that have bound self-antigen. To begin characterizing the molecular requirements for B cell migration into follicles, cells were treated with pertussis toxin (PTX), an inhibitor of signaling by many G protein-coupled chemokine receptors. Lymphocyte accumulation in t...

Lymphocyte Migration: A new spin on lymphocyte homing

New evidence confirms the multi-step model for leukocyte migration through endothelia, but with a twist. The selectins are not always required, and some lymphocyte subsets rely heavily on integrins for their migration to tissues.

Cell Migration in the Immune System: the Evolving Inter-Related Roles of Adhesion Molecules and Proteinases

Leukocyte extravasation into perivascular tissue during inflammation and lymphocyte homing to lymphoid organs involve transient adhesion to the vessel endothelium, followed by transmigration through the endothelial cell (EC) layer and establishment of residency at the tissue site for a period of time. In these processes, leukocytes undergo multiple attachments to, and detachments from, the vess...

Essential role of Elmo1 in Dock2-dependent lymphocyte migration.

Elmo1 and Elmo2 are highly homologous cytoplasmic adaptor proteins that interact with Dock family guanine nucleotide exchange factors to promote activation of the small GTPase Rac. In T lymphocytes, Dock2 is essential for CCR7- and CXCR4-dependent Rac activation and chemotaxis, but the role of Elmo proteins in regulating Dock2 function in primary T cells is not known. In this article, we show t...
